10.4.: Gastvortrag: The modern single life: stochastic modelling of individual cells and molecules

Abstract:
The recent introduction of experimental techniques with single molecule and single cell resolution have challenged traditional views of molecular mechanisms in biology; processes such as transcription have been found to be subject to substantial amounts of stochastic variation. It is unclear what sources of this ‘noise’ are and how cells cope with it. I will focus on our recent developments of superior experimental techniques and modelling approaches to allow novel insights into mammalian transcription. This will include the application of next generation sequencing to single cells, as well as the combination of flow cytometry with fluorescence in situ hybridization. We further use Bayesian statistical frameworks to interpret the experimental data and fit models to it. These interdisciplinary approaches allow us to understand transcriptional dynamics on both, a genome-wide scale, as well as on the single molecule and single cell level.
